21CFR11 COMPLIANCE AND GLP VALIDATION

With the security and electronic records features in Version 14, the TOX IVOS is fully compliant with 21CFR11. Specifically developed to comply with regulatory guidelines, the TOX IVOS II is supported by full documentation. Our TOX IVOS II Validation Handbook outlines specific GLP protocols to verify the accuracy of internal calculations. CUSTOMIZED ALGORITHMS FOR RAT SPERM ANALYSIS

Because of the large size of the rat sperm and the unique shape of its head, customized algorithms have been developed for rat sperm analysis with the TOX IVOS II. Using these special algorithms, the TOX IVOS II digitally “separates” the sperm head from the midpiece. The TOX IVOS II is able to isolate the sperm head in this method because it is the only sperm analyzer that uses the whole sperm image, not just the border, for analysis. The precise identification and tracking of sperm can be immediately confirmed with the fulldisclosure Playback feature of the TOX IVOS II.

ACCURATE TOTAL SPERM COUNTS IN HOMOGENIZED TISSUE

The time-consuming task of hemacytometer sperm counts is now a thing of the past. For total sperm counts in homogenized tissue, nothing matches the ease of use and accuracy of the IDENT fluorescing system. Integrated into the TOX IVOS II, the IDENT fluorescent illumination is activated at the touch of a button. Sample preparation for the IDENT analysis requires a simple three minute staining procedure. Since the IDENT stain is DNA-specific, sperm heads are clearly illuminated and identified while debris is ignored. As with standard analysis, accurate identification is immediately confirmed with the Playback feature.

AUTOMATIC STORAGE OF DIGITAL IMAGES

For compliance with regulatory guidelines, raw data of all fields analyzed may be automatically saved to hard drive, external hard drive or internal network with the Digital Image Storage Option. Because the exact sperm images are saved, digital image storage is the optimum QC method. For easy retrieval, the analysis setup, the study name and the animal number are stored with each image file.

INTEGRATED OPTICS

As the only sperm analysis system with an internal optical system, the TOX IVOS II offers distinct advantages to the sperm analysis laboratory. In contrast to the continuous illumination of a microscope, the TOX IVOS uses illumination strobed at 1/1000 of a second to visualize sperm motion. This strobed illumination eliminates motion-related blurring along the length of the sperm head, resulting in precise sperm tracking. By adding an image capture rate of 60 frames per second, you get the highest level of accuracy available today for measuring sperm velocities and motion parameters. For added flexibility, the TOX IVOS II performs analyses under three types of internal, strobed illumination: phase contrast, bright field and multi-wavelength fluorescence.

AUTOMATED, INTERNAL, HEATED SPECIMEN STAGE

As part of the integrated optical system, the unique computer controlled specimen stage of the TOX IVOS II provides precise control of temperature and position during analysis. The stage temperature, which may be set from ambient to 40 °C, remains constant to within 0.5 °C. For selection of analysis fields, the stage may be programmed for either manual or automatic field selection. For at-a-glance monitoring, the current stage temperature and position are continuously shown on screen as a real-time, digital display. The IVOS stage accommodates and automatically adjusts for commonly used analysis chambers, such as disposable fixed-depth slides, cannulas and Makler chambers. In addition, a user-defined chamber setting is available.
